I am trying to develop a software for autocollimator to detect shift in pixels and then convert that shift into arc seconds reading

I am able to interface the camera but i want to know what image processing techniques i should use to 1)for filtering
2)for image data processing for detecting centroid of the reflected crossline from autocollimator
